چکیده

The bacteriostatic action of 2-chloro-4-aminobenzoic acid (2-Cl-PAB) was found by Wyss, Rubin, and Strandskov (1943) to be completely reversed by p-aminobenzoic acid. This, according to the theory of Woods (1940) that p-aminobenzoic acid is a specific antagonist for sulfanilamide, classified the bacteriostatic action of 2-Cl-PAB as sulfonamide activity. The bacteriostatic action of this compound did, however, differ from that of the sulfonamides in that it could be completely reversed by methionine. The action of the sulfonamides is only partially reversed by this amino acid. Shive and Roberts (1946) hav-e suggested that the sulfonamides interfere wvith a number of enzyme systems in which p-aminobenzoic acid functions and that the enzyme system which is most sensitive to the sulfonamides is the one responsible for the synthesis of methionine. Because the bacteriostatic action of 2-Cl-PAB is completely revTersed by methionine, they postulate that 2-Cl-PAB inhibits only the enzyme system involved in the synthesis of this amino acid. The work reported here is a study of the inhibition of methionine synthesis bv 2-Cl-PAB and sulfanilamide. Strains of Escherichia coli made resistant to 2-Cl-PAB and a strain made resistant to sulfanilamide were employed. The mechanism of the development of resistance to 2-Cl-PAB was also investigated.

The replacement of para-aminobenzoic acid by methionine in the growth of a Neurospora mutant.

Present knowledge of the function of para-aminobenzoic acid (PABA) has been derived almost exclusively from sulfanilamide (SAN) inhibition studies. Shive and Roberts (1946) have advanced the hypothesis that PABA is catalytically active in the synthesis of a number of metabolic products in Escherichia coli and that the enzyme systems concerned in their production are differentially sensitive to ...

Methionine reversal of 2-chloro-4-aminobenzoic acid inhibition in Escherichia coli.

During recent years many experiments have demonstrated the usefulness of antimetabolites for studying routes of metabolism. Competitive inhibitors have been used to block specific enzyme systems and to establish enzyme-substrate relationships, and the compounds produced by the action of these enzyme systems have been characterized through their ability to enhance the reversal of the inhibitors ...

Studies on the accumulation of 4-amino-5-imidazole carboxamide in Escherichia coli.

When cultures of Escherichia coli are partially inhibited by sulfonamides they accumulate a diazotizable and nonacetylable amine in the medium (Fox, 1942). The amine was isolated by Stetten and Fox (1945) and characterized by Shive et al. (1947) as 4-amino-5-imidazole carboxamide. The latter authors suggested that the amine was a precursor of purines as it only required the addition of a one ca...

Inhibition Steps in Sulfonamide Bacteriostasis of Escherichia Coli.

The effectiveness of p-aminobenzoic acid (PABA) in overcoming sulfanilamide (SA) growth inhibition of Escherichia coli was explained on the basis that PABA was an essential metabolite and that SA interfered with -its utilization by the cell (Woods, 1940). Reversal of sulfonamide action by structurally unrelated metabolites such as methionine, xanthine, serine, thymine and valine (Shive, 1950; W...
